Fresno, CA doesn't have an interstate running through it (I-5 is maybe 50 miles away). There are several cities larger than Greenville in CA, and others elsewhere, that don't have interstate access in the city.
It does surprise me that Greenville doesn't have a interstate. Wilson even has two (I-95 & I-795). In my opinion they should've made 795 go from 95 to Greenville instead of going to Goldsboro.
But for Fresno not to have a interstate is a little ridiculous, doesn't the city have close 500,000 people?

Fresno is just over 500k, with a metro area around 1m.
I haven't found anything showing total eastern NC population, and I don't feel like adding up the population totals of the roughly 40 counties (I assume that's what you're wondering).

Wilson doesn't actually have any. Neither of those Interstates were built to service Wilson. Wilson just so happens to be where the Interstates went.
Goldsboro got the Interstate because of the air force base.
The Greenville-New Bern-Washington DMA has 595,100 people in it.

University Health Systems uses the number 1.3 million. So I'll use that number. And that doesn't include the southeast.

This past Sunday I was saying to my wife that the southeast portion of Greenville was very underserved by any auto parts stores. It turns out to be a "Ask and Ye shall receive" type thing. I was on Firetower driving past the strip shopping center where Michealangelos Pizza is and there was a sign announcing a new O'Reilly's Auto Parts was opening soon.
The McDonalds close to greenville blvd. and 10th street has been completely bulldozed.
McDonald's usually completely rebuilds its current locations rather than abandon and relocate.
