I am 55 and single and just got offered a good job in Manhattan. Living in NYC has always been a fantasy of mine. Can a middle-aged single woman meet people and have a good time in New York, or did I miss my chance and Manhattan is only for young single women?

I am not sure what your life situation is but I think there are a lot middle aged single men here too .
I don't think NYC has an age limit. Althought sometimes I wish things had an age minimum because I cannot stand children.
all of my parents' friends bought homes in manhattan and brooklyn and they're well past middle age - they love it! its a lot more convenient to shop, eat out or go to the theatre
my parents refuse to take the plunge though

Take the job- it is a great experience to live in NY !
But be prepared, it not easy to make friends here. Single women are afraid of competition and act accordingly, single men from 25 to 75 years old are looking for women in their twenties.
I think Manhattan is the ideal place for a single woman.
You don't need a car so you don't have to worry that there is no "man" around to fix it or deal with an auto mechanic
You can dine at a nice restaurant all by yourself if you want to and no one will blink and no one will bother you.
If you want to go to a movie, concert or the theater and you have no one to accompany you, you can go by yourself without it looking awkward.
If you have any medical problems or develop any new ones as you age, you have tons and tons of options for medical care.
You don't have to worry about managing to walk home with bags of heavy groceries to feed a family because you only have to feed yourself.
There are tons of lectures, classes, groups, wine tastings, clubs on a wide variety of topics and areas of interest where you can meet new people.
I think Manhattan is the hardest for little kids who are often cooped up in a small place and have no place to run around outside freely without an adult hovering.
Try meetup.com...I keep posting this even though I haven't attended too many, but it seems like a fabulous way to meet like minded people no matter what your interests may be. Some events are targeted for singles, but many more are not. Only caveat is that a lot of them seem geared toward younger folks, but not all.

Sure, just watch out for the late 20 - 30 something year old bitter singles that will call you a birther regardless if if you had kids or not.
I've been wanting to do the same thing and I'm in my 40's. I think you should do it!
