It's an easy drive, one that I've personally done at least 3 times. Asheville is a fantastic town, but I have to assume you've been, and it doesn't shave a whole lot of time off for you. The last few exits in NC before TN are places I would not recommend at all to stop for the night. I can't remember the exact town it was, but I was told by the hotel manager there that "they wash their sheets."

Yeah. I just sort of assumed that was done at all hotels. apparently not in that town.
If you're going to do it over two days, I'd suggest leaving early, getting through the smokies, and staying somewhere around Knoxville. It isn't a hard drive though, just load up some podcasts, leave early, and get to Nashville in the evening, even if you take it easy.
