I made it to the second phase interview with the pharmacist and store manager...just waiting for them to schedule same...said they would call after Tuesday.
You may be among other candidates that have moved onto the next phase of the interview process. The initial phase should have weeded out the worst candidates. The upcoming phase will determine which candidate is the best to be hired.
Noticing the position is a cashier/pharmacy technician trainee, being a registered or state certified technician in your state, successfully completing a pharmacy technician education program, and/or having worked in a pharmacy elsewhere would be necessary or helpful in being hired. (CVS does have an Accreditation Council for Pharmacy Education-approved pharmacy technician program.)
It's only a good sign when you get an official offer letter with terms of acceptance and information such as starting salary, that you sign, date, and return.

None of us can tell you for sure.

C'mon guys, it's a retail cashier/tech job. What letter, they don't do that for that type of position. You know you're hired when you pass the drug test and come in to fill out your paperwork. Until then don't assume.
OP being sent for a drug test is a good sign, at that point they have a good enough feeling about you to spend money for the test.
Cashier and pharmacy technician are two separate positions. Pharmacy technicians are involved in the fill process of prescriptions, and must be registered or state certified with their state's board of pharmacy. Cashiers operate registers.
The original poster is likely being considered for a pharmacy cashier position.

Decades past, I didn't have to interview for a cashier position at a Kmart, let alone go through a 2nd round of interviews. AFAIK, that may have changed.

Interviews are pretty standard now, and only if you manage to pass the online app first. I Imagine the first interview was with store mgr or asst to see if candidate was suitable per store expectations. The second round will likely be in conjunction with Rx mgr to get more into the specific requirements of working in the pharmacy, as that position usually requires a candidate to have even better people skills (and thick skin helps too, lol).
I interviewed for a support/inventory/floor position for a store ten years ago and was interviewed by a committee of managers from three different stores, I thought it was a bit of overkill myself.
