Just saw this article on CNN. Looks like Raleigh was one of the big "winners" for the 2010 Census. Population has grown 41% since 2000, 2nd only to Provo Utah. I know a lot of people have differing feelings about the population growth here, but at least it should mean more federal funding.
The Census Bureau does sampling throughout the decade. They may not have precise numbers, but with well-developed sampling techniques, they (and other population experts) can predict with a reasonable degree of accuracy.
There is a whole demographics department within the state, full of mathematicians who are constantly analyzing population changes in the state, regardless of the decennial Census (I interviewed to work for them once--would have been a dream job ).
