NE-71, from Crawford to the South Dakota line. It was REALLY lonely 20 years ago when it was still a gravel road, especially at night.
CA-62, from Vidal Junction to Twentynine Palms.
TX-207, between Claude and Silverton, but well worth the trip when you suddenly drop off into Palo Duro Canyon. Yes, it really IS a 10% grade! If you happen to be driving a fully loaded 18 wheeler, as I was the first time I went that way, it'll make a believer out of you real fast.
CO-109. from La Junta to the junction with US-160 (which is pretty damn lonely itself from Springfield to Trinidad). Don't be surprised if you meet a B-52 about 100 ft off the ground. That'll wake you up!
MT-13, from Circle up to Wolf Point. There MAY be 3 farmhouses along that route. Try it in a blizzard sometime. At night.
TX-54, from Guadalupe Peak to Van Horn. There's one ranch house and one pipeline pump station. That's it.
M-28 acrost upper Michigan. The "seney stretch" is famous for being isolated. ( just west of Newberry, all the way to Munising) Much of the trip is empty swamp, pine forrest, a great place to fall asleep and run off the road.
The US-54 going from Emporia, KS to Santa Rosa, NM.
I've heard of the US-50, but plenty of my old trucking trips had me using the 54 on more than one occasion.
I remember having to layover in Minneola, KS for a 24 period to pass in order to wait for 10 hrs to fall off my logbook time.
I-10 after San Antonio till you get to El Paso. Oh about 9 hrs worth traveling 80mph. Most states you can drive through in less than that. You do not want to break down or get a flat. There's stretches of I-10 that has nothing, not even a gas station for 50-100 miles easy.
