How did Comcast determine that I may have malware on a computer in my home?
We identify infected computers in several ways. First, we get data from reputable Internet research groups that specialize in bot identification. This data includes a list of Internet Protocol (IP) addresses that are infected or belong to bot command and control channels. Second, we look for malicious behavior exhibited by bots such as sending spam, distributed denial of service attacks and repeated connection requests to known command and control channels. We then aggregate this data to confirm that behavior typically associated with malware was detected.

Basically, Comcast watches the network traffic and can determine from that if you may have a bot on your computer doing something malicious. From what I've read, the scan that Comcast has you run may not be all that effective in actually getting rid of it. Most people suggest running one of two programs:
1. Malwarebytes - It's a free download and a really good program and will get rid of almost anything.
2. SpyBot Search & Destroy - Another freebie that is good that can also help protect your computer upfront.
If you have Windows7, you should be running MSE (Microsoft Security Essentials), it is a great program and since I've been using it I haven't gotten any infections that it didn't immediately detect and eliminate. Before that I was running the two programs above on my old XP machine.
