There is noting in the Constitution about political parties at all. In an general election we vote for the candidate not the party.
Election laws are by state and there are rules in each state stating how to get on the ballot. The only way to get someone off the ballot is to show to a judge that the person did not qualify to be on the ballot according to the laws of the state. Parties typically don't do this but opposing candidates do and it can be members from the same party(in an primary election) or opposing parties(in a general election against 3rd parties). A third party candidate could indeed be too weak to something like gather enough valid signatures to be put on the ballot.
